    Why Use Chicken IgY?

  • Here are top 5 reasons:

    1. Higher titres against highly conserved mammalian gene products

    2. Double immunostaining is easier to perform

    3. Animal-friendly -- we purify the antibodies from eggs, not serum

    4. Large quantities of antibody - faster and cheaper

    5. Nearly unlimited quantities (again, because the antibodies come from eggs)

Amyloid Precursor Protein (APP), Peptide #5

Price: $399
Catalog #: APP5
Data Sheet (PDF)
Mouse cortical neurons in culture. The green staining is APP-immunoreactivity, using fluorescein-labeled goat anti-chicken IgY (Product #F-1005, Aves Labs) and rhodamine-labeled phalloidin as a counterstain. Note the APP-staining of the neurites and growth cones, and the phalloidin-staining limited to the distal growth cones and filapodia. Photomicrograph courtesy of Dr. Philip Copenhaver (OHSU).

Product Info

Antibody Concentrations:  This antibody preparation [Amyloid Precursor Protein (APP), Peptide #5] is at a concentration of 100 ug/ml (based on absorbance at 280 nm).

Volume: Regular vials contain 1000 µl of this antibody mixture; Sampler vials contain 200 µl of this mixture.

Buffer: Phosphate-buffered (10 mM) isotonic (0.9%, w/v) saline (PBS, pH 7.2) with sodium azide (0.02%, w/v) added as a preservative.

Quality Control:  Antibodies were validated by immunohistochemistry (at a dilution of 1:1000) using fluorescein-labeled goat anti-chicken IgY (1:500 dilution, Aves Labs Cat.# F-1005) as the secondary reagent.

Recommended Dilutions: 1:500-1:1000 for immunohistochemistry and immunocytochemistry using 2% paraformaldehyde-fixed tissues or cells. Note that these dilutions are meant to serve as starting points, and that optimal dilutions may vary.

Description: Chickens were immunized with synthetic peptide APP#5 conjugated to keyhole limpet hemocyanin (KLH).  The sequence of APP peptide #5 is EEI QDE VDE LLQ KEQ NYS DD, which corresponds to residues #556-575 of the human gene product (NP_000475.1)].  Identical sequences are also found in the mouse and rat homologs of APP.  After repeated injections, immune eggs were collected, and the IgY fractions were purified from the yolks.  These IgY fractions were then affinity-purified using a peptide column, and the concentrations of the eluates adjusted to 100 ug/ml.  Finally, equal volumes of these affinity-purified anti-peptide antibodies were mixed, and the preparation was filter-sterilized.
