Chickens were immunized with a synthetic peptide containing the human c-myc epitope tag – EQKLISEEDL – conjugated to keyhole limpet hemocyanin (KLH). The sequence of this peptide corresponds to residues #425-434 of the human c-myc proto-oncogene protein (NCBI NP_002458.2). After repeated injections, immune eggs were collected from the hens, and the IgY fractions were purified from the yolks. These IgY fractions were then affinity-purified using an agarose matrix to which the cognate peptide was attached. Eluates of this column were dialyzed against PBS, and then conjugated to fluorescein using its isothiocyanate form (FITC). Fluorescein-labeled antibody was separated from free FITC using gel exclusion chromatography (Sephadex G-50), and the protein concentration was adjusted to 0.1 mg/mL in PBS with 0.02% sodium azide. Finally, the preparation was filter-sterilized.

Store at 4°C in the dark. Under these conditions, the antibodies should have a shelf life of at least 12 months (provided they remain sterile). For longer storage periods, dilute the antibodies with sterile glycerol and store at -20°C. Do not freeze these antibodies unless you want to store them for longer periods of time.
